Necroptosis in tumorigenesis, activation of anti-tumor immunity, and cancer therapy
While the mechanisms underlying apoptosis and autophagy have been well characterized over recent decades, another regulated cell death event, necroptosis, remains poorly understood. Elucidating the signaling networks involved in the regulation of necroptosis may allow this form of regulated cell death to be exploited for diagnosis and treatment of cancer, and will contribute to the understandin...
متن کاملRUNX3 methylation and anti-tumor immunity.
Altered DNA methylation is a hallmark of cancer genomes and has been advocated as both a marker and mechanism of the malignant phenotype. In the conventional view, such alterations include global DNA hypomethylation, linked to aberrant expression of oncogenic driver genes and more localised de novo hypermethylation at ‘CpG-island’ promoters, argued to cause inappropriate silencing of tumor supp...
متن کاملEngineering Anti-Tumor T Cell Immunity
T cell immunity is critical for protection against infectious agents as well as cancer. T cell immune response is a well orchestrated process that involves three key components. CD8+ T cells that harbor cytolytic machinery and can target and kill the tumor cells in an antigen specific manner, CD4+ T cells that can either “help” the generation of a productive CD8+ T cell or “regulate/suppress” i...
متن کاملHost CD73 impairs anti-tumor immunity
The enzymatic activity of CD73 produces immune-suppressing adenosine. In CD73 deficient hosts, tumor growth and tumor infiltration by Tregs and type 2 immunosuppressive macrophages is reduced. Pharmacological inhibition of CD73 in wild-type mice has similar tumor-suppressing effects. Host CD73 on leukocytes and endothelial cells is thus detrimental for the anti-tumor immunity.
متن کاملAn evolutionary perspective on anti-tumor immunity
The challenges associated with demonstrating a durable response using molecular-targeted therapies in cancer has sparked a renewed interest in viewing cancer from an evolutionary perspective. Evolutionary processes have three common traits: heterogeneity, dynamics, and a selective fitness landscape. Mutagens randomly alter the genome of host cells creating a population of cells that contain dif...
